Description

The dry version of the broth we make in our own lab, in one pouch. Myco Labs Culture Blend is a pre-measured mix of extra light malt extract, dextrose, soy peptone and our nutritional supplement and mineral blend. Weigh it, dissolve it in hot water, and pressure cook. One 250 g pouch makes over 3 US gallons of liquid culture.

Why growers use Culture Blend

How to make liquid culture

  1. Mix — add the measured Culture Blend to the matching amount of hot or boiling water and stir until fully dissolved.
  2. Filter (optional) — pour the mixture through a paper coffee filter to reduce light malt extract sediment.
  3. Cool and fill — let the broth cool, then fill your jars, leaving headspace at the top.
  4. Sterilize — pressure cook for 30 minutes at 15 PSI.
  5. Cool, then inoculate — once the jars are back to room temperature, add 3–5 cc of spore solution or liquid culture, or a clean agar wedge.
  6. Incubate at 76–82°F, out of direct light, and swirl or stir regularly to keep growth even.

Expect an amber broth. Culture Blend is malt-based, so finished broth is yellow to light amber rather than water-clear. That is normal. Keep to the 30-minute cook time; running much longer can darken the sugars.

How much liquid culture does one pouch make?

One 250 g pouch makes over 3 US gallons of liquid culture broth at the label ratio. That works out to about 12 quart batches or 25 pint batches.

How much Culture Blend do I use per jar?

Measure by weight: 9.7 g per US pint of water, 19.4 g per US quart, or 77.5 g per US gallon. In metric that is about 20.5 g per liter. Use a gram scale rather than a measuring spoon so every batch comes out the same.

What is in Culture Blend?

Extra light malt extract, dextrose, soy peptone, and a proprietary nutritional supplement and mineral blend. It is the dry version of the broth recipe our lab has refined over 15 years of producing liquid cultures.

What color should the finished broth be?

Yellow to light amber. The color comes from the malt extract, so the broth will not be water-clear, and that is normal. It stays transparent enough to see mycelium growing in the jar.

Do I have to filter the broth?

No, filtering is optional. Pouring the dissolved mix through a paper coffee filter before you fill your jars removes light malt extract sediment, which makes new growth easier to spot and keeps the culture easier to draw through a needle.

How long do I sterilize it?

Pressure cook for 30 minutes at 15 PSI. Cooking much longer than you need to can darken the sugars in the broth, so stick to the time on the label, then let the jars cool completely to room temperature before you inoculate.

What can I inoculate the broth with?

Spore solution, an existing liquid culture, or a clean agar wedge. For a jar, 3 to 5 cc of spore solution or liquid culture is all it takes.

How should I store the dry mix?

Keep the pouch tightly sealed in a cool, dry place and always use a clean, dry utensil when measuring. Each pouch carries an expiration date on the back. Culture Blend is for mushroom cultivation and is not for human consumption.
